FITC-LC-Peptid anstelle von FITC-Peptid
Wir empfehlen bei der Auswahl von Fluorescein (FITC) als N-terminalem Fluoreszenzlabel dieses mit einem "long chain (C6 spacer / Ahx [ 6-amino caproic acid / 6-amino hexanoic acid]" von der nachfolgenden Peptidsequenz zu trennen.
Das Label "FITC" zeigt, im Gegensatz zum Label "FAM" die Tendenz, sich während der gewünschten Markierungsreaktion der Peptide gemeinsam mit der N-terminalen Aminosäure abzuspalten. Vergleiche hierzu : Edman Degradation.
Der bei der Peptidkonjugation mit dem FITC-Farbstoff unerwünschte Degradationsvorgang kann durch einfügen eines LC Spacers zwischen FITC und Peptidsequenz weitesgehend vermieden werden.
